Breaking New Ground in Alzheimer's Care: With Only 32 Patients

Challenge

ANAVEX®2-73 (blarcamesine), a Sigma-1 receptor agonist developed by Anavex Life Sciences, needed validated biomarkers to identify which Alzheimer's patients would respond to treatment : starting from a small Phase I-II cohort, with no established precision-medicine pathway for the drug yet in place.

Approach

KEM® analyzed complex, heterogeneous molecular data - including genetic, microbiota, and real-world evidence data - from a 32-patient Phase IIa cohort. Biomarkers identified in this analysis were subsequently confirmed in an open-label extension study.

Results

ANAVEX®2-73 showed a statistically significant reduction in disease progression over a 148-week trial period. Patients in the high-concentration cohort showed an 88% improvement over the low-concentration cohort in maintaining performance in activities of daily living. Further genomic analysis confirmed the SIGMAR1 gene variant as a key driver of treatment efficacy. This precision-medicine approach also contributed to Anavex's market valuation rising from $150M to over $1B.

Business Outcome

The biomarker findings opened three new potential indications - Rett Syndrome, Dementia, and Parkinson's Disease - supported by regulatory milestones including Fast Track designation. Anavex's market valuation rose from $150M to over $1B following the study.
